    Protective effect of Nigella sativa oil on acoustic trauma induced hearing loss in rats

    Acoustic trauma is a common reason for hearing loss. Different agents are used to prevent the harmful effect of acoustic trauma on hearing. The aim of this study was to evaluate the potential preventive effect of Nigella sativa (black cumin) oil in acoustic trauma. Our experimental study was conducted with 20 Sprague Downey female rats (mean age, 12 months; mean weight 250 g). All of the procedures were held under general anesthesia. Following otoscopic examinations, baseline-hearing thresholds were obtained using auditory brainstem responses (ABR). To create acoustic trauma, the rats were then exposed to white band noise of 4 kHz with an intensity level of 107 dB in a soundproof testing room. On Day 1 following acoustic trauma, hearing threshold measurements were repeated. The rats were divided into two groups as the study group (n: 10) and the controls (n: 10). 2 mL/kg/day of Nigella sativa oil was given to the rats in the study group orally. On Day 4 following acoustic trauma, ABR measurements were repeated again. There was no difference between the baseline hearing thresholds of the rats before acoustic trauma (P>0.005). After the acoustic trauma, hearing thresholds were increased and there was no significant statistically difference between the hearing thresholds of the study and control groups (P=0.979). At the 4th day following acoustic trauma, hearing thresholds of the rats in control group were found to be higher than those in the study group (P=0.03). Our results suggest that Nigella sativa oil has a protective effect against acoustic trauma in early period. This finding should be supported with additional experimental and clinical studies, especially to determine the optimal dose, duration and frequency of potential Nigella sativa oil therapy.

    Ratlarda akustik travma sonrası çörek otu yağı uygulamasının elektrofizyolojik etkilerinin araştırılması (Deneysel Çalışma)

    Akustik travma sık karşılaşılan işitme kaybı nedenlerindendir. Tedavisinde farklı ajanlar kullanılmasıyla birlikte fikir birliğine varılamamıştır. Çalışmamızın amacı çörek otu yağının akustik travmada etkilerini Auditory Brainstem Response “İşitsel Beyin Sapı Cevapları” yöntemini kullanarak değerlendirmektir. Çalışmamız 20 adet yaş ortalaması 12 ay ve ortalama ağırlıkları 250 gr olan Sprague Downey cinsi dişi rat üzerinde deneysel araştırma olarak tamamlanmıştır. Ratlar genel anestezi altında otoskopik muayeneleri ve ABR testleri yapılarak akustik travma öncesi işitme eşikleri saptanmıştır. Daha sonra sessiz kabinde 12 saat süre ile 4 kHz’de 107 dB şiddetinde beyaz bant gürültü verilerek akustik travma yaratıldı. Travma sonrası 1. gün ABR ile işitme eşikleri tekrar ölçüldü. Ratlar her bir grupta 10 adet olmak üzere ilaç ve kontrol grubu olmak üzere ikiye ayrıldı. 4. gün ölçümler tekrarlandı işitme eşikleri iki grup arasında karşılaştırıldı. Akustik travma öncesi yapılan testler sonucunda tüm ratların eşikleri benzer olarak bulundu (p> 0,005). Akustik travma sonrasındaki ilk ölçümlerde tüm ratlarda eşiklerin yükseldiği, her iki grupta eşikler arasında istatistiksel fark olmadığı saptandı (p=0,979). Çörek otu uygulamasının ardından 4. Günde yapılan ölçümlerde ise kontrol grubunun işitme eşiklerinin çalışma grubuna göre daha yüksek olduğu saptandı (p=0,03). Çalışmamızın sonucunda çörek otu yağının kullanımının akustik travmada yararlı olabileceği saptanmıştır. Bu bulgular ışığında çörek otu kullanımının dozu, süresi ve uygulama sıklığı açısından yapılacak ileri çalışmalara ihtiyaç vardır. Acoustic trauma is a common reason for hearing loss. In post trauma day 1; ABR test results and hearing thresholds were measured. Then rats were divided into two groups; the study group (n: 10, nigella sativa oil) and the control group (n: 10). On post trauma day 4; ABR retest was performed again and results were compared. Prior to the acoustics trauma, hearing threshold results of the the rats were similar (p> 0,005). After the acoustic trauma, hearing thresholds was increased and there was no significant statistically difference between the thresholds between two groups (p=0,979). After nigella sativa oil implementation of 4th day, hearing thresholds of control group was higher than the study group (p=0, 03). It was found that nigella sativa oil might have a positive effect against acoustic trauma. However, it is further studies concerning nigella sativa oil usage dose, period and sequence are needed.
