Item Association of Plasma GDF-9 or GDF-15 Levels with Bone Parameters in Polycystic Ovary Syndrome(2015) Berberoglu, Zehra; Aktas, Aynur; Fidan, Yasemin; Yazici, Ayse Canan; Aral, Yalcin; 0000-0002-3132-242X; 24430093; AAS-6810-2021We aimed to determine plasma levels of growth and differentiation factor (GDF)-9 and GDF-15, and their possible association with bone turnover parameters and bone mineral density (BMD), in patients with polycystic ovary syndrome (PCOS). Forty-two obese PCOS women aged 25-35 years, 23 women with idiopathic hirsutism (IH) and 20 healthy controls matched for age and body mass index were enrolled. Anthropometric, metabolic and hormonal patterns, plasma GDF-9 and GDF-15 concentrations, bone turnover markers and BMD were measured. No significant differences were observed in bone turnover markers, BMD measurements, plasma GDF-9 and GDF-15 levels in subjects with PCOS compared with the other two groups. In the combined population of all three groups, GDF-15 concentrations were negatively correlated with osteocalcin (r = -0.317, p < 0.01). Analysis of PCOS patients showed a significant correlation of GDF-15 concentrations with age and homeostasis model assessment index (r = 0.319, p < 0.05, and r = 0.312, p < 0.05, respectively). In addition, GDF-15 concentrations were negatively correlated with osteocalcin (r = -0.395, p < 0.01) and positively correlated with urine deoxypyridinoline (r = 0.353, p < 0.05). GDF-9 did not correlate with bone markers and BMD measurements. In conclusion, plasma GDF-9 and GDF-15 levels as well as bone turnover markers and BMD measurements in subjects with PCOS (25-35 years of age) were comparable with those either in subjects with IH or in healthy controls with similar anthropometric and metabolic profiles. We aimed to determine plasma levels of growth differentiation factor (GDF)-15 and their possible association with hormonal and metabolic status, as well as echocardiographic profiles and carotid artery intima-media thickness (CAIMT) measurements in patients with polycystic ovary syndrome (PCOS). Methods. Forty-two obese PCOS women aged 25-35 years, 23 women with idiopathic hirsutism and 20 healthy controls matched for age and body mass index were enrolled. Anthropometric, metabolic and hormonal patterns, plasma GDF-15 concentrations, CAIMT, and conventional echocardiographic parameters were measured. Results. Metabolic/lipid profiles as well as GDF-15 levels were similar across the three groups. CAIMT tended to be higher in PCOS group but did not reach statistical significance. No between-group differences were found in the conventional echocardiographic parameters. Analysis of PCOS patients showed a significant correlation of GDF-15 concentrations with age and homeostasis model assessment (HOMA) index (r=0.319, P<0.05, and r=0.312, P<0.05, respectively). In multiple linear regression analyses, GDF-15 was significantly associated with age (r(2)=0.102, P<0.05), and HOMA index (r(2)=0.10, P<0.05). Conclusion. Plasma GDF-15 levels, CAIMT and conventional echocardiographic parameters in obese subjects with PCOS (25-35 yrs old) were comparable to those in either subjects with idiopathic hirsutism or healthy controls with similar anthropometric and metabolic profiles, suggesting that PCOS alone could not impart an early and higher risk independent of associated risk factors. We aimed to investigate the role of serum betatrophin in full-blown polycystic ovary syndrome (PCOS) patients and 47-aged healthy women, 51 full-blown PCOS patients were included in this cross-sectional study. Betatrophin concentrations were significantly lower in PCOS group and displayed a positive correlation only with serum tryglyceride in control group (p < .05). A cutoff level (464.5 ng/L) was determined for betatrophin according to Receiver Operating Characteristic curve. Using this value, 64.7% of PCOS patients were classified as below the cutoff and in this group betatrophin was found to correlate negatively with fasting glucose, fasting insulin, and homeostasis model assessment of insulin resistance (p = .038, p = .020, and p = .014, respectively), and positively with total testosterone (p = .041). In the rest of PCOS cases (35.3%) who had betatrophin higher than cutoff, positive correlation was found with low-density lipoprotein cholesterol (p = .009). In conclusion, betatrophin levels are reduced in full-blown PCOS patients who had worse metabolic phenotype.Item Adenovirus-36 Infection and Obesity: A Case Control Study of Turkish Women with Polycystic Ovary Syndrome(2021) Cakmakliogullari, Elcin Kal; Eroglu, Semra; 0000-0002-9959-9433; 33581595Background and aims: Adenovirus-36 (Ad-36) seropositivity has been shown to be involved in the aetiology of obesity. The aim of this study was to examine Ad-36 positivity in obese and normal-weight patients with polycystic ovary syndrome (PCOS). Methods: There were two groups including 92 and 110 subjects. This study was a prospective casecontrol study. The enzyme-immunoassay method was used to quantitatively determine antibodies (Abs) specific to human Ad-36 in the serum samples. Age, body mass index (BMI), fasting glucose levels and insulin levels of the participants were recorded. The PCOS and control group patients were divided into two groups: the overweight group with BMI >= 25 kg/m(2) and non-obese group with BMI <25 kg/m(2). Results: Ad-36 Ab positivity in the PCOS group was found to be significantly higher than that in the control group (p < 0.001). Ad-36 Ab positivity was significantly higher in the PCOS obese group than in the control obese group (p < 0.001). Ad-36 Ab positivity and BMI >= 25 kg/m(2) were identified as independent predictors of PCOS in logistic regression analysis. Conclusion: Ad-36 Ab positivity was significantly higher in the obese/overweight PCOS patients. Obesity can be prevented in patients with PCOS by treating Ad-36. (C) 2021 Diabetes India. This parasite plays a role in the pathophysiology of acne in polycystic ovarian syndrome (PCOS). We aimed to examine the relationship between Demodex folliculorum and blood glucose control in patients with PCOS with skin and eyelash lesions. Forty-four patients with PCOS with skin lesions were enrolled in the study. At least two specimens were taken from the skin lesions using the standard method and at least six epilated eyelashes were taken from both eyes under a biomicroscope and evaluated using a light microscope. The demographic characteristics, body mass index (BMI) and clinical parameters of the patients were recorded. Demodex folliculorum was present at a rate of 59.1% in the skin lesions of the patients with PCOS, 40.9% in eyelash samples and 43.18% in both skin and lashes. Homeostasis model assessment of insulin resistance (HOMA-IR), glycosylated haemoglobin (HB A1c) concentrations, and BMIs were significantly higher in the patients who had D. folliculorum in skin samples than in those without (p = .010, p = .007 and p = .02). Impaired glucose regulation may explain the pathophysiology of the increased D. folliculorum presence in the skin lesions of patients with PCOS.Impact statement What is already known on this subject? Although several studies on Demodex folliculorum and PCOS have been conducted, we have not yet found a study that examines D. folliculorum parasites in the eyelashes and skin correlating with glucose regulation in PCOS. This study presents new information about the relationship between the presence of D. folliculorum and impaired glucose regulation in women with PCOS. What do the results of this study add? D. folliculorum is seen more commonly in skin lesions in patients with PCOS with impaired blood glucose regulation.
