Item Optimization Of Spray-Drying Process Parameters For Microencapsulation Of Three Probiotic Lactic Acid Bacteria Selected By Their High Viability Rate In Sucrose And Fructose Levels And High Temperatures(Başketn Üniversitesi Kazan Meslek Yüksekokulu, 2024-05-05) Bagdat, Elif Seyma; Akman, Perihan Kubra; Kutlu, Gozde; Tornuk, FatihMicroencapsulation is an efficient way to increase the survival rate of probiotics against harsh conditions. In this study, three probiotic strains (Lactiplantibacillus plantarum subsp. plantarum strain W2 (LP4), Lactiplantibacillus pentosus strain XL640 (LPE1), and Limosilactobacillus fermentum strain W8 (LF2)), isolated from shalgam and gilaburu, were microencapsulated with spray drying and process conditions [maltodextrin concentration (MC, 10-30%) and inlet air temperature (IAT, 110-130 degrees C)] were optimized by central composite rotatable design of response surface methodology. The results indicated that the predicted IAT and MC values for the maximum powder yield and viability were 123.21 degrees C and 22.76%, 130.37 degrees C and 19.49%, and 127.94 degrees C and 10.00% for LF2, LP4 and LPE1, respectively. At these conditions, bacterial viability ranged from 10.27 to 10.33 log colony-forming units per gram (cfu/g), while the powder yield values for the encapsulation of the bacteria were between 43.38% and 50.97%. Furthermore, MC was the most significant factor for the powder yield of LF2, LPE1, and viability of LPE1. Encapsulation efficiency values higher than 92.77% demonstrated the efficiency of spray drying for the protection of selected strains. The microcapsules produced at the optimum points had moisture content between 5.30 and 5.96%. SEM images showed that the microcapsules were in spherical shape. In conclusion, the results confirmed that the selected probiotics were successfully microencapsulated with spray drying with high powder yield, viability, and encapsulation efficiency levels and these features could reveal the potential of the encapsulated probiotic strains to be used in high-sugar foods.Item Esra (Elevator Simulation, Research & Analysis): An Open-Source Software Tool For Elevator Traffic Simulation, Research, And Analysis(Başkent Üniversitesi Teknik Bilimler Meslek Yüksekokulu, 2024-04-05) Tartan, Emre Oner; Ciflikli, CebrailToday, although elevator companies typically have their own elevator simulation and analysis tools, these remain inaccessible to the public due to commercial concerns and rivalry in the sector. Consequently, due to the lack of modular open-source software tools, interest in the design and development of elevator systems in academia is quite limited, contrary to the vast potential of the field. This paper addresses this gap introducing an open-source software tool, the Elevator Simulator for Research and Analysis (ESRA), developed using the MATLAB programming language and an object-oriented approach. ESRA features built-in control methods, and visualization options with the capability to save and reuse traffic data, facilitating in-depth comparative analysis. Moreover, its modular architecture allows a design and development basis for further research in the field. G.; Bengi, S.; Durmus, P.; Demirezen, S.; Altindal, S.The capacitance/conductance-voltage-frequency (C/G-V-f) features of the Au/p-Si structures with (%0.5 Bi-doped ZnO) interlayer grown by spin-coating technique was investigated between 100 kHz and 1 MHz. The C-2-V curves that depend on frequency were used to determine the electrical-parameters like diffusion-potential (V-D), Fermi-energy level (E-F), barrier-height (Phi(B)), and depletion-layer (W-D) thickness. The value of Phi(B) was found to increase with increasing frequency due to the decrease in surface-states (N-ss) effects on the C-V characteristics. Voltage dependent series-resistance (R-s) was obtained by using Nicollian-Brews method for each frequency and it decreased with increasing frequency. Both the low-high frequency capacitance (C-LF-C-HF) and Hill-Coleman techniques were used to determine the voltage-dependent profile of N-ss. While N-ss decreases with increasing frequency, voltage-dependent on them give two distinctive peaks due to a special distribution of them at interlayer/semiconductor interface in the bandgap depending on their relaxation time. All these experimental results indicate that the existence of R-s, N-ss, and interlayers considerably influence the impedance measurements.Item Comparison Of Efficacy And Reliability Of Six Commercial Covid-19 Diagnostic PCR Kits(Başkent Üniversitesi Kazan Meslek Yüksek Okulu, 2024-03-26) Sharafi, Parisa; Gokdemir, F. All countries have been deeply affected by the coronavirus disease 2019 pandemic, both economically and in situations that strain health systems, such as workforce and workload. Therefore, various measures should be taken to control the disease and prevent its spread. Since the disease onset, real-time PCR tests have been used as the gold standard for disease diagnosis. Owing to the rapid progress of the pandemic and the spread of the disease, validation, consistency, and optimization tests of some commercial kits have been conducted directly in the field. Therefore, it is important to compare the results of these kits and improve the existing ones. Methods. We compared five kits (Bioexen, Polgen, Coronex, Diagen, and Anatolia) donated to the TOBB Economics and Technology University Hospital PCR laboratory with the KrosGen kit to detect severe acute respiratory syndrome coronavirus 2. A total of 244 samples were selected and analyzed using five different severe acute respiratory syndrome coronavirus 2 PCR detection kits. Results. Positive and negative results from the six kits were compared using the working protocols of the kits, primers, and cycle threshold (Ct) values. Five of the six kits have reliable compatibility for Ct<30 but decreases for Ct >= 30. Therefore, it is important to evaluate the performance of these kits for reduced viral loads. Conclusions. Using a suitable kit with high compatibility for Ct >= 30 is important for detecting patients with a low viral load and helping prevent disease spread.Item Evaluating The Multifaceted Impact Of Cephalaria Extract On Dough Quality: Antioxidant, Antimicrobial, And Cytotoxic Properties(INTERNATIONAL JOURNAL OF FOOD SCIENCE AND TECHNOLOGY, 2024-12) Bagdat, Elif Seyma; Bozkurt, Fatih; Ilikkan, Ozge Kahraman; Sagdic, Osman; Tornuk, FatihCephalaria syriaca, known as cephalaria, is an annual, pilose plant with pink-purple flowers growing wild in wheat fields. Viscoelastic properties of gluten-free doughs can be improved by adding cephalaria. However, this addition results in some undesirable properties in bread such as bitter taste and dark internal colour. This study aimed to reduce these effects by obtaining cephalaria obtained from distilled water and ethanol extracts and to increase the potential use of extracts in bread production. Different proportions of water and ethanol extracts obtained from oil-free cephalaria were added to bread by creating a model with Mixture Design, and the optimum concentration was determined by farinograph trials. The antioxidative, antimicrobial, and cytotoxic effects of extract and the effect of cephalaria on yeast activity were examined. Cephalaria extracts had no measurable antimicrobial effect and no antifungal effect on Rhizopus stolonifer, while an antifungal effect was found against A. niger and P. expansum. The IC50 value was found to be 4.15 mg mL(-1 )for ethanol extract while the water extract had no effect on cells. The addition of extract had no negative effect on the number of yeasts in sourdough fermentation. The study has improved the usage of cephalaria by using water and ethanol extracts in dough.Item A Novel Synbiotic Edible Film Based On Aquafaba, Psyllium Husk Powder, Peg 400, And Lactiplantibacillus Plantarum 299v And Applicability On Kashar Cheese(JOURNAL OF FOOD MEASUREMENT AND CHARACTERIZATION, 2024-12) Ilikkan, Ozge Kahraman; Bagdat, Elif Seyma; Yilmaz, Ilkay; Beyter, Nurten; Yuzay, Isinay EbruIn this study, it was aimed to compare a novel synbiotic edible film based on aquafaba, psyllium husk powder, and PEG 400 with or without Lactiplantibacillus plantarum 299v. Therefore, primarily, optimization of the edible film was carried out by Box-Behnken design in response surface methodology (rsm). The characterization of edible films with or without probiotic bacteria was performed by FTIR, viscosity, pH, total solid content, water and oxygen solubility, film thickness, color L*, a*, b*, transparency, and film tensile strength. It was determined that there was no statistically significant difference in viscosity, pH, lightness L*, water vapor, and oxygen permeability values with the addition of probiotic lactic acid bacteria to the optimized coating film samples. In addition, it was observed that the probiotic bacteria count did not decrease below 108 cfu/g for 60 days of storage. The resulting edible film formulation with (PL) and without (PS) probiotics was used to coat Kashar cheese, while control cheeses (C) were not coated. As a result, it was observed that the microbiological quality of the coated cheeses was as good as control or better than control especially at the end of storage in terms of mold/ yeast and coliforms. Sensory evolution revealed coating films with or without probiotics did not change the texture, flavor, odor, or appearance of cheeses. Physicochemical properties such as pH, % titratable acidity, % dry matter, % fat, and % salt were detected.Item The Effect Of Organizational Culture İn Health Services On Employee Flourishing(WORK-A JOURNAL OF PREVENTION ASSESSMENT & REHABILITATION, 2024-11-15) Cagatay, Huseyin Tolga; Demirel, Yuksel; Ertugrul, Bekir; Tekinyildiz, EsraBACKGROUND: Culture has an important role in maximizing the value of human capital, which is regarded as the main source of organizational effectiveness. OBJECTIVE: The research was conducted to determine the effect of organizational culture perceptions of healthcare professionals working in a university hospital on employee flourishing. METHODS: It is a descriptive study conducted with 299 healthcare professionals working in a university hospital. "Structured Personal Information Form", "Denison Organizational Culture Survey" and "New Flourishing Index" to collect the data. In the analysis descriptive statistical methods such as percentage, frequency, arithmetic mean and standard deviation as well as Pearson correlation, linear regression and multiple linear regression analyses were performed to determine the relationship between variables. RESULTS: According to the results of linear regression analysis, it was determined that organizational culture and flourishing were related (F: 88.229; p < 0.05). The organizational culture alone explained 22.6% of flourishing. As a result of multiple regression analysis, it was determined that the relationship between organizational culture sub-dimensions and flourishing was significant (F = 25.975; p < 0.05). The sub-dimensions of organizational culture explained 25.1% of the flourishing. CONCLUSIONS: As a result of the study, it is observed that the perception of organizational culture of healthcare professionals has an impact on flourishing, and therefore the favorable development of organizational culture will contribute positively to employee flourishing. In present conditions, it has become a necessity for health organizations to provide the circumstances that will make all investments that will contribute positively to the flourishing of their employees a part of the organizational culture.Item Anaesthetic Management for Orthotopic Liver Transplantation in A Patient with Glycogen Storage Disease Type Iiia(2023) Haka, D.; Cekmen, N.; 0000-0001-7448-8203Glycogen storage disease (GSD) type III is a metabolic disorder caused by a deficiency in amylo-1,6-glucosidase enzyme, which is responsible for the breakdown of the glycogen molecule, resulting in glycogen accumulating in the organs, hypoglycaemia, muscle weakness, liver dysfunction, delayed anaesthetic recovery, excessive surgical bleeding, cardiomyopathy and end-organ dysfunction. This case report presents a child with GSD type IIIa who underwent orthotopic liver transplantation (OLT) with her mother as a donor. A multidisciplinary approach should be provided to optimise the preoperative period and minimise complications in these patients.Item Sequential Decision Making for Elevator Control(2023) Tartan, Emre Oner; Ciflikli, Cebrail; 0000-0002-5688-4226; JVD-9650-2023In the last decade Reinforcement Learning (RL) has significantly changed the conventional control paradigm in many fields. RL approach is spreading with many applications such as autonomous driving and industry automation. Markov Decision Process (MDP) forms a mathematical idealized basis for RL if the explicit model is available. Dynamic programming allows to find an optimal policy for sequential decision making in a MDP. In this study we consider the elevator control as a sequential decision making problem, describe it as a MDP with finite state space and solve it using dynamic programming. At each decision making time step we aim to take the optimal action to minimize the total of hall call waiting times in the episodic task. We consider a sample 6-floor building and simulate the proposed method in comparison with the conventional Nearest Car Method (NCM).
