Item The Effect of Thyroid Dysfunction on Nesfatin-1 And Adiponectin Levels in Rats(2017) Atici, Emine; Mogulkoc, Rasim; Baltaci, Abdulkerim Kasim; Menevse, Esma; 0000-0003-2461-1212; 0000-0002-5477-5667; 0000-0002-6547-4798; 28796641; AAL-5444-2020; ABA-7831-2020; AAG-5865-2019Background: Changes in thyroid hormone concentrations may affect adiponectin concentrations through various mechanisms. A molecule released primarily from the fat cells adiposities; adiponectin has important effects on the regulation of body weight. Aim: The present study aimed to explore the effects of experimental thyroid dysfunction and its treatment on nesfatin-1 and adiponectin levels in rats. Methods: The study included 40 adult male Sprague-Dawley rats which were grouped as follows: (1) control; (2) hypothyroidism [hypothyroidism was induced by intraperitoneal injection of 10 mg/kg/day propylthiouracil (PTU) for 3 weeks]; (3) hypothyroidism + thyroxine group [after hypothyroidism was induced by 2-week PTU injection, they were treated with high-dose L-thyroxine (1.5 mg/kg/day) for 1 week]; (4) hyperthyroidism [hyperthyroidism was induced by 3-weeks' thyroxine injection (0.3 mg/kg/day)]; (5) hyperthyroidism + PTU (after hyperthyroidism was induced by 2-weeks' thyroxine injection, the animals were given 10 mg/kg/day PTU for 1 week). Blood samples taken at the end of the study were analyzed to measure nesfatin-1 and adiponectin levels. Results: It was found that nesfatin-1 levels increased in hypothyroidism, while adiponectin levels decreased (p < 0.001). In experimental hyperthyroidism, on the other hand, both nesfatin-1 and adiponectin levels were found significantly elevated (p < 0.001). Conclusion: The results of the study indicate that nesfatin-1 and adiponectin levels were modified considerably in hypo- and hyperthyroidism, whereas with the restoration of the thyroid function, modified hormone levels went back to normal.Item Co-Culture of Rat Luteal Cells with Islet Cells Enhances Islet Viability and Revascularization(2018) Boyuk, Gulbahar; Yigit, A. Arzu; Aydogan, Ilkay; 0000-0001-5837-6877; 30187177; AAD-1857-2020; C-2636-2017Islet cell transplantation is a major treatment strategy for type I diabetes, and has proven to be effective for maintaining glucose homeostasis. However, this treatment requires an extended period of immunosuppression to prevent rejection and recurrent transplantation to maintain function. Thus, to enhance the properties of transplanted islet cells, we examined the effect of the co-culture of luteal cells, which secrete progesterone, on islet cell viability, functionality, and revascularization. It was found that islet viability and functionality were higher in the co-cultured group than in single cultures of islets at 48 and 96 h, in parallel with increased progesterone and vascular endothelial growth factor (VEGF) secretion from luteal cells. In the co-culture groups, VEGF levels at 48 and 96 h and CD31 levels at 48 h were significantly higher than those in the islet groups (p < 0.001 and p < 0.05, respectively), and basic fibroblast growth factor (bFGF) levels were increased at 96 h (p < 0.001). Thus, co-culture with luteal cells may increase islet vascularity by enhancing VEGF and bFGF levels for up to 96 h, which could help to markedly increase the pre-transplantation time to allow for effective immunosuppression therapy. This method may also promote islet cell viability and functionality. Progesterone and angiogenic factors secreted from luteal cells may be responsible for these positive effects.Item Resonant frequency shifts related to the elastic constants near the alpha-beta transition in quartz(2019) Yurtseven, H.; Ates, S.The resonant frequency shifts of vibrational modes are related to the elastic constants at various temperatures for the alpha-beta transition in quartz (T-c = 573.0 degrees C for alpha-quartz and 574.3 degrees C for beta-quartz). This correlation is constructed on the basis of the spectroscopic modifications of the Pippard relations near the alpha-beta transition in quartz by using the observed data from the literature. Our results indicate that the elastic properties can be described from the spectroscopic parameters (frequency shifts) as we have studied here for the alpha-beta transition in quartz. Using those relations, elastic constants can also be obtained from the frequency shifts near the phase transitions of various crystalline systems. (C) 2018 Elsevier B.V. Methods: 62 volunteer soccer players from Cayyolu Sports Club were included in our study. Players were randomly allocated into two groups; a Kinesio taping (KT) (n=31), and a control group (n = 31). KT application including gastrocnemius and quadriceps femoris muscle facilitation techniques were applied to the first group. The second group was set as the control group and no application was made. Sociodemographic characteristics, postural stability and thermographic assessment were evaluated prior to and after the application of the Kinesio Taping. All the statistical analyses were set a priori at an alpha level of p<0.05. The tests for homogeneity (Levene's test) and normality (Shapiro-Wilk) were used to determine the appropriate statistical methods to apply for comparison between the groups. According to the test results, the parametric paired sample T-test was used to compare between baseline and post-treatment within a group, while independent sample T-test for equality of means was used for comparison between the groups. Results: No statistically significant differences were observed between the groups at baseline regarding age, body mass index, skin temperature and postural stability (p > 0.05). Statistically significant differences were found in all the assessed parameters in both Kinesio Taping and control groups (all p<0.05) except thermal imaging of dominant gastrocnemius muscle results (p=0.668) after the Kinesio application. Statistically significant differences were found in all parameters across groups (all p<0.05). Conclusions: Kinesio Taping may increase postural stability and decrease the risk of injury. According to thermal imaging assessments, both quadriceps and gastrocnemius muscles may be active in maintaining postural stability. As the decreasing risk of injury is very important for young soccer players, our results may show a preventive measure for sports physiotherapists working on soccer fields.Item Value of cardiopulmonary exercise testing in the diagnosis of coronary artery disease(2019) Akinci Ozyurek, Berna; Savas Bozbas, Serife; Aydinalp, Alp; Bozbas, Huseyin; Ulubay, Gaye; 31414640Introduction: Respiratory and cardiac functions in association with skeletal and neurophysiologic systems can be evaluated with cardiopulmonary exercise testing (CPET). Compared to treadmill exercise test, CPET provides more comprehensive data about the hemodynamic response to exercise. Materials and Methods: We aimed to evaluate the relationship with CPET findings and coronary lesions identified on angiography in patients with angina pectoris who underwent teradmill exercise, CPET and coronary angiography (CAG). By this way we sought to examine the CPET parameters that might be predictive for coronary artery disease (CAD) before diagnostic exercise test results and ischemia symptoms develop. Thirty patients in whom CAG was planned because of symptoms and exercise test results were enrolled in the study. Oxygen consumption (VO2), carbon dioxide production (VCO2), minute ventilation (VE), maximum work rate (WR), Delta VO2/Delta WR and O-2 pulse (VO2/HR) values were calculated. Significant CAD was defined as >= 50% narrowing in at least one of the coronary arteries. Results: The mean age was 60.4 +/- 8.9 years ve 21 (65.6%) of subjects were male. On CAG, CAD was detected in 19 (59.4%) patients. Maximum heart rate, heart rate reserve (HRR), VE/VCO2 measured at anaerobic threshold AT) and VO2 (mL/kg/min) were significantly differed in patients with CAD than those without (p= 0.031; p= 0.041; p= 0.028; p= 0.03 respectively). Peak VO2, VO2/WR and O-2 pulse values were higher in patients with normal angiographic results than those with CAD but the difference did not reach to statistical significance. Conclusion: The findings of our study indicate that among CPET parameters AT VE/VCO2, ATVO(2) (mL/kg/dk) and HRR can have predictive value in the diagnosis of CAD. We think that these parameters might be used in the evaluation of patients with angina and dyspnea suspected of CAD. In conclusion parameters obtained during the test that are not influenced by patient's effort might increase the value of CPET in the diagnosis CAD.Item HEV seroprevalence in blood donors in Turkey by two commercial total anti-HEV Ab ELISA kits(2019) Yasar, Osman; Karatayli, Ersin; Cengiz, Guniz; Kizilpinar, Mehtap; Yurdcu, Esra; Albayrak, Rabia; Guven, Aysel; Arslan, Onder; Karahan, Ceren; Otlu, Baris; Guducuoglu, Huseyin; Gokahmetoglu, Selma; Berk, Elife; Gitmisoglu, Ozlem; Sertoz, Ruchan; Yurdaydin, Cihan; Bozdayi, A. Mithat; Karatayli, Senem Ceren; 31403185Previous hepatitis E virus (HEV) seroprevalence studies in Turkey have shown high variabilities, leading to conflicting results. We aimed to re-evaluate HEV seroprevalence among blood donors in Turkey using the Wantai (Beijing, China) and the Dia.Pro (Milan, Italy) total anti-HEV antibody (Ab) enzyme-linked immunosorbent assay (ELISA) kits and compare their performances and to investigate the presence of HEV RNA in blood donors. Serum total anti-HEV antibodies were determined in a total of 2011 volunteer blood donor samples collected from different regions of Turkey (807 from Ankara, 243 from Kayseri, 284 from Izmir, 200 from Malatya, 200 from Kahramanmaras, and 277 from Van). HEV RNA was evaluated by a real-time polymerase chain reaction in a total of 272 anti-HEV seropositive samples. The country-wide HEV seroprevalence was calculated as 11.5% (Dia.Pro) and 12.2% (Wantai) with seropositivity rates of 12.0%-12.5% in Ankara, 7.4%-8.2% in Kayseri, 14.5%-15.5% in Malatya, 8.1%-8.8% in Izmir, 15.0%-16.0% in Kahramanmaras, and 12.6%-13.4% in Van by Dia.Pro and Wantai kits, respectively. The lowest detectable Ab concentrations were 0.16 and 0.14 units/mL WHO, for the Dia.Pro and the Wantai assays, respectively, showing no significant difference between assays. HEV RNA was not detected in any of the anti-HEV seropositive samples. Compared with previous studies, HEV was shown to have a higher overall seroprevalence in Turkey. Despite its limitation, the current study represents the most comprehensive HEV seroprevalence study in Turkey performed with two different commercial ELISA assays with high sensitivities so far. Further investigation is required to determine HEV genotypes in Turkey.Item EFFECT OF GREEN TEA (CAMELLIA SINENSIS L.) AND PARSLEY (PETROSELINUM CRISPUM) DIETS AGAINST CARBON TETRACHLORIDE HEPATOTOXICITY IN ALBINO MICE(2019) Guven, Aysel; Nur, Gokhan; Deveci, Haci AhmetIn this study, it has been tried to be searched the effects of green tea (Camellia sinensis L.) and parsley (Petroselinum crispum) that is known their antioxidant features on levels of MDA, GSH and activities of GSH-Px, GST, CAT on mice liver tissue. In the study, total 32 Swiss albino mice has been used that 8 of them are control and 24 of them are practise. Group I: animals were put on a normal diet and sham-treated with 2 ml/kg distilled water through oral gavage, daily for 8 weeks; this group of animals served as the control. Group II: animals were put on a normal diet and treated with 1.5 ml/kg b.w CCl4 dissolved in 1.5 ml distilled water through oral gavage. Group III: animals were put on a normal diet and treated with 1.5 ml/kg b.w CCl4 + 500 mg/kg b.w green tea through oral gavage. Group IV: animals were put on a normal diet and treated with and for CCl4 + 0,464 g/kg parsley oral gavage daily for 8 weeks. As a results, by increasing antioxidant enzymes levels on groups given green tea and parsley to control group (p<0,05), but in MDA levels has been seen a significant decreasing as statistical (p<0.01). While both Green tea and Parsley were found to have a protective effect against CCl4 induced damage, Parsley was more protective. Histopathological analysis revealed that liver tissue appeared normal in control on the other hand, there were degeneration, congestion, cellular infiltration, and necrotic areas in the group administered with CCl4. Even though frequency of lesions decreased, similar lesions were observed in the group with 1,5 ml/kg b.w CCl4. Consequently, it was determined that while CCl4 administration increased oxidative stress, green tea and parsley administration had a protective potential increasing antioxidant capacity. Natural antioxidant substances found in parsley and green tea can be considered as the best green chemical substances to cope with oxidative stress without damaging the nature and the living.Item Evaluation of long-term effects of diode laser application in periodontal treatment of poorly controlled type 2 diabetic patients with chronic periodontitis(2019) Dengizek Eltas, Seydanur; Gursel, Mihtikar; Eltas, Abubekir; Alptekin, Nilgun Ozlem; Ataoglu, Tamer; 0000-0003-4104-6462; 30697968; G-1816-2014Purpose This study aimed to investigate the effects of diode laser (DL) in addition to non-surgical periodontal treatment on periodontal parameters, systemic inflammatory response and serum haemoglobin A(1c) (HbA(1c)) level in patients with poorly controlled type 2 diabetes mellitus (T2DM) and chronic periodontitis. Methods Thirty-seven patients with poorly controlled T2DM and chronic periodontitis completed this study. The patients were divided into two groups. The individuals in the control group received placebo laser treatment in addition to scaling and root planing (SRP). The individuals in the study group received DL (1 watt) in addition to SRP. Clinical index measurements were performed before treatment (T0), 3 months after treatment (T1) and 6 months after treatment (T2). Plaque index, gingival index, bleeding on probing, clinical attachment level and probing depth were measured to determine periodontal status. HbA1(c) and C-reactive protein (CRP) levels were also analysed using blood samples. Results In both groups, clinical and laboratory parameters were significantly improved at T1 and T2 compared to baseline (P < 0.05). Gingival index, bleeding on probing and probing depth were more significantly reduced after treatment in the SRP+DL group than in the SRP group (P < 0.05). The serum CRP and HbA(1c) levels were similar between the groups (P > 0.05). Conclusion The use of DL in addition to SRP in periodontal treatment of T2DM individuals makes positive contribution to the reduction of local inflammation and to periodontal healing. On the other hand, it has no beneficial effects on systemic inflammatory response and glycaemic control.Item Periodontal status and dental behaviour of heart transplant recipients and healthy controls(2019) Sezgin, Yasemin; Sezgin, Atila; 31661343; AAC-5931-2020Objective: The aim of this study was to compare the periodontal status and dental behaviour of heart transplant recipients with systemically healthy controls. Material and Methods: Forty heart transplant recipients and 40 systemically healthy patients were included. A questionnaire was used to record demographic, systemic health and dental behaviour data, and the following clinical parameters were recorded: plaque index, bleeding on probing, probing depth, gingival recession, and clinical attachment level. Results: Mean probing depth (p = .045), mean gingival recession (p = .016), mean clinical attachment level (p = .021) and mean plaque index scores (p = .001) were higher in heart transplant recipients than the systemically healthy group. However, bleeding on probing was similar in both groups (p > .05). Tooth brushing frequency and number of participants who used dental floss/interdental brush was lower in the heart transplant group. Conclusion: Considering the poor oral hygiene and high clinical periodontal parameters of heart transplant recipients, periodontal evaluation should be done regularly before and after transplantation. The patient's daily oral hygiene regimens should be evaluated carefully in this evaluation session and modified based on their gingival health, manual skill and motivational levels. The importance of oral health and its effects on systemic health should also be explained to the transplant recipients in detail.Item Efficacy of ultrasound-guided bilateral erector spinae plane block in pediatric laparoscopic cholecystectomy: Case series(2019) Karaca, Omer; Pinar, Huseyin Ulas; 0000-0003-0473-6763; 31741347; Q-2420-2015Postoperative opioid administration can cause various side effects, such as drowsiness, respiratory distress, postoperative nausea, and vomiting. The use of non-opioid medications as part of a multimodal analgesia method has been increasingly suggested in the management of acute postsurgical pain. Erector spinae plane block (ESPB), which is a regional anesthesia technique, blocks both visceral and somatic nerve fibers.Though it is infrequently used in young patients, presently described is a series of cases in which ESPB was successfully used in the performance of pediatric laparoscopic cholecystectomy. Ultrasound-guided bilateral ESPB was performed on 4 patients who underwent a laparoscopic cholecystectomy. An injection of 0.25% bupivacaine was administered into the interfascial area on each side in the pre-incisional period after the anesthesia induction (total anesthetic: 2.5 mg/kg). Postoperative pain control was planned at 10 mg/kg intravenous paracetamol every 8 hours. Numeric rating scale pain scores were less than 3 points in the post-anesthetic care unit, and at 1, 2, 4, 8, 12, and 24 hours. No rescue analgesic (1 mg/kg tramadol) was needed. None of the typical complications, such as drowsiness or nausea, were observed and no block-related complications were recorded. Bilateral ESPB that is a part of multimodal analgesia regimen can provide effective analgesia for pediatric laparoscopic cholecystectomy patients in the first 24 hours postoperatively.
