    Protective Effect of Spirulina on Cisplatin-Induced Ototoxicity: A Functional and Histopathological Study

    Objective: The purpose of this study was to evaluate the protective effect of an antioxidant and anti-inflammatory agent, "spirulina," against cisplatin-induced ototoxicity in rats. Methods: Twenty-eight adult Sprague-Dawley rats were divided into 4 groups. Before drug administration, distortion product otoacoustic emission and auditory brainstem response tests were performed. Group 1 (n =7) received 1 mg of intraperitoneal saline. Group 2 (n=7) received a single dose of intraperitoneal cisplatin at 15 mg/kg/day. Group 3 (n=7) received oral spirulina at 1000 mg/kg/day for 10 days. Group 4 (n=7) received a single i.p. dose of cisplatin at 15 mg/kg/day, followed by 10 days of oral spirulina at 1000 mg/kg/day. The final distortion product otoacoustic emission and auditory brainstem response measurements were provided 10 days after the initial drug administration. Cochleas were removed, the histochemical examination was performed by caspase-3, caspase-9, and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling methods. Results: Initially, there were no significant differences in distortion product otoacoustic emission and auditory brainstem response measurements between groups. Following cisplatin treatment, the mean difference in signal to noise ratio values was lower in the cisplatin + spirulina group compared to the cisplatin-only group. The increase in auditory brainstem response thresholds was more significant in the cisplatin-only group than in the cisplatin + spirulina group. Posttreatment auditory brainstem response latencies were prolonged in cisplatin and cisplatin + spirulina groups; however, a significant difference was obtained between these 2 groups. The cisplatin + spirulina group had a lower density of apoptotic cells than the cisplatin-only group. Conclusion: Spirulina has no adverse effects on cochlear functions and may provide some protection against cisplatin-induced ototoxicity.

    Spirilunanın ratlarda sisplatin ototoksisitesi üzerine etkisi

    Amaç: Antioksidan etkinliği bilinen spirulinanın sisplatin ototoksitesine karşı koruyucu etkisinin olup olmadığını değerlendirmektir. Gereç ve Yöntem: Prospektif kontrollü bu hayvan çalışmasında 2 8 adet Sprague Dawley rat dört gruba ayrıldı. İlaç uygulamalarından önce ve 10 günlük ilaç uygulaması sonrasında her bir gruptaki ratların her iki kulağına distorsiyon ürünü otoakustik emisyon testi (DPOAE) ve işitsel beyin sapı yanıtı (ABR) testleri yapıldı. 1. Gruptaki ratlara (n = 7), tek doz 1 ml intraperitoneal serum fizyolojik (%0.9) verilirken, 2. gruptaki ratlara (n = 7 ), 15 mg/kg/gün tek doz sisplatin intraperitoneal olarak verildi. 3 . gruptaki ratlara (n = 7), 1000 mg/kg/gün spiriluna 10 gün süreyle gavaj ile verildi. 4. gruptaki ratlara (n = 7) 15 mg/kg/gün şekildeki tek doz sisplatin uygulaması ile aynı gün başlanan 1000 mg/kg/gün spirulina 10 gün süreyle gavaj ile verildi. Bulgular: Çalışma öncesi yapılan ölçümlerde gruplar arasında herhangi bir anlamlı fark bulunmadı. DPOAE sonuçları açısından 1. grupta ve 3 . grupta ilaç uygulaması öncesi ve sonrasında herhangi bir frekansta anlamlı bir fark gözlenemedi. 2. Grupta ise 2222 Hz, 2500 Hz, 3200 Hz, 4444 Hz, 5000 Hz, 8000 Hz, 8889 Hz, 10000 Hz ve 11429 Hz frekanslarında ilaç uygulaması sonrası SNR değerleri uygulama öncesine göre daha düşük bulundu. 4. Grupta 4444 Hz, 5 000 Hz, 8000 Hz, 8 8 89 Hz, 10000 Hz ve 11429 Hz frekanslarında ilaç uygulaması sonrası SNR değerleri uygulama öncesine göre daha düşük bulundu. Sadece sisplatin alan ratlar diğer gruplar ile karşılaştırıldığında daha düşük SNR değerleri elde edildi. Son ölçümlerde 1.grup 3 ve 4. Grup ile ikili olarak karşılaştırıldığında aralarında anlamlı fark gözlenmedi. İlaç uygulaması öncesinde grupların ABR eşikleri arasında anlamlı fark bulunmaz iken ilaç uygulaması sonrası 4.grubun ABR eşiklerinin 2. gruba göre daha iyi görüldü. Sadece sisplatin alan ratlarda ise diğer gruplara göre ABR yanıtı daha yüksek eşik değerlerinde elde edilebildi. Before the drug administration, distortion product otoacoustic emission (DPOAE) and Auditory Brainstem Response (ABR) tests were performed. Group 1 (n = 7), received 1 mg intraperitoenal (i.p.) saline. Group 2 (n = 7) received single dose of intraperitoenal (i.p.) cisplatin at 15 mg/kg/day. Group 3 (n = 7) received oral spirulina at 1000 mg/kg/day for 10 consecutive days. Group 4 (n = 7) single dose of intraperitoenal (i.p.) cisplatin at 15 mg/kg/day follwed by oral spirulina at 1000 mg/kg/day for 10 consecutive days. Final DPOAE and ABR measures were performed 10 days after the first drug administration. Results: Before the durg administration there was no significant differences between groups in terms of DPOAE and ABR measurements. Group 1 and 3 showed no significant difference before the drug administration and DPOAE measurements before. Group 2 demonstared lower SNR values at 2222 Hz, 2500 Hz, 3200 Hz, 4444 Hz, 5000 Hz, 8000 Hz, 8889 Hz, 10000 Hz ve 11429 Hz frequencies in final DPOAE measurement. In group 4 final DPOAE SNR values were lower in 4444 Hz, 5000 Hz, 8000 Hz, 8889 Hz, 10000 Hz ve 11429 Hz frequencies.In pairwise comparisons; group 1, 3 and 4 had no significat difference in final DPOAE measurement however rats received only cisplatin had lower SNR values than in other groups. There was no significant difference in terms of ABR threshold before the durg administration. After the drug administration, increase in ABR treshols was greater in group 2 than in group 4. Rats received only cisplatin yielded higher ABR tresholds than rats in other groups Conclusion: Spirulina does not have negative effect on cochlear functions and has a partial protective role against cisplatin-induced ototoxicity.
