Item Maternal ve fetal MBL2 genotiplerinin preterm doğumlarla ilişkisi(Başkent Üniversitesi Sağlık Bilimleri Enstitüsü, 2009) Taneri, Ayşe; Ataç, F. BelginYenidoğan döneminde, immün sistemin tam olarak gelismemesine bağlı karsılasılan komplikasyonlara ek olarak, yenidoğanın “prematüre” olması prenatal mortalite ve morbidite hızını arttıran baslıca faktördür. Görülme sıklığının %9 olması nedeni ile, günümüz perinatal tıbbının çözmeye çalıstığı ve çevresel, tıbbi ve kalıtsal faktörlerin rol oynadığı çoklu değisimlere bağlı bir yenidoğan sorunudur. Patogenezi aydınlatmaya yönelik yapılan çalısmalarda inflamasyonun gestasyonel süreci etkilemesi, preterm etkeni olarak kabul görmesine neden olmustur. Bu bulgu inflamatuar yanıtın olusmasında rol oynayan proteinleri kodlayan genlerdeki polimorfizmlerin, preterm yatkınlık faktörü olarak arastırılmasını gerektirmektedir. Mannoz bağlayan lektin-2 (MBL2) karaciğerden sentezlenir ve hipogammaglobulinemik fazda rol oynayan bir lektin yolağı proteinidir. Maternal serum düzeyinin gebeliğin ilk trimesterinde artısına ek olarak nidasyon, plasentasyon ve hamileliğin devamında islevsel olduğunun anlasılması, MBL2’nin gebelik komplikasyonları-inflamasyon iliskisinin aydınlatılmasında belirteç olabileceğini göstermistir. Serum düzey farklılığı, 10q11.2-q21’e lokalize olan MBL2 geninin promotör ve 1. ekzonundaki tek nükleotid polimorfizmlerinden (SNP) kaynaklanmaktadır. Promotör bölgedeki polimorfizmler genin transkripsiyonunu etkileyerek protein düzeyindeki değisimlerden sorumludur. MBL geninin 1. ekzonunda meydana gelen gen varyantları ise proteinin homopolimer özelliğini bozarak, sitoplazmada degredasyona yatkın hale gelmesine neden olur. Bu bilgiler doğrultusunda, çalısmamızda term ve preterm doğumlarla maternal–fetal MBL2 ekzon 1 genotiplerinin iliskisi arastırılmıstır. Bu amaçla, 100 term (38,8±1,2 hafta) ve 83 pretermden (30,2±2,7 hafta) elde edilen maternal ve kord kan örneklerinden genomik DNA izolasyonu yapılmıstır. Daha sonra MBL2 kodon 52 (db SNP ID rs5030737), 54 (db SNP ID rs1800450) ve 57 (db SNP ID rs1800451) genotiplemesi PZR-RFLP analizi ile gerçeklestirilmistir. Kodon 57 polimorfizminin toplumumuza özgü olmadığı gözlenirken, term doğumlarda maternal kodon 54 230 G/A genotipinin preterm doğum yapan annelere göre anlamlı derecede yüksek olduğu (p<0.045) saptanmıstır. Preterm grupta, fetal-maternal kodon 54 GG genotip sıklığının term fetal-maternal gruba göre daha yüksek olduğu saptanmıstır (p<0.001). Gebelik inflamasyonun arttığı bir süreçtir. Proinflamatuar sitokin düzeyindeki artıs gestasyonel süreci de kısaltır. Önceki çalısmalarda, kodon 54 230 G/A genotipinin düsük düzeyde MBL2 sentezlediği gösterilmistir. Bu veriler birlestirildiğinde, gestasyonel sürecin uzamasına neden olan genotipin bir avantaj sağladığı görülmektedir. Multifaktöriyel hastalıklarda rol oynayan mekanizmaların tek bir parametre ile açıklanması söz konusu değildir. Gestasyonel sürecin devamlılığının sağlanması için fetal-maternal immünitenin karsılıklı regülasyonunda rol oynayan genler ile ilgili veri tabanlarının olusturulması, ile risk gruplarının belirlenmesi mümkün olabilecektir. Bu nedenle, bu çalısmanın fetal-maternal immün regülasyonda rolü olan diğer faktörlerle birlestirilerek devam etmesi gerektiği kanısına varılmıstır. In addition to complications associated with the immature immune system of newborns, “preterm” birth is the main factor that increases prenatal mortality and morbidity. With a 9% incidence, it is a newborn problem associated with multi factorial variations, which environmental, medical and genetic factors enroll that has been tried to be solved. The molecular studies strongly indicate inflammation as a risk factor for shorter gestational age. These findings strongly suggest that gene polymorphisms in the inflammatory pathway are a candidate for the explanation to preterm tendency. A lectin pathway component mannose binding lectin-2 (MBL2) is synthesized in liver and participates in hypogammaglobulinemic phase. MBL2 is assigned as a marker for the explanation of pregnancy complications and inflammation not only due the increase in serum level in the first trimester of pregnancy but also it participates in nidation, placentation and maintenance of pregnancy. MBL2 gene is located at 10q11.2-q21. The single nucleotide polymorphisms (SNPs) in promoter and/or in the first exon of the relative gene are responsible for the interindividual serum MBL level. Polymorphisms in promoter are responsible from changes in protein level by affecting transcription rate, where as variants in exon 1 are the cause of susceptibility to degradation in cytoplasm by degrading homopolymer structure of protein. Therefore, we felt it was prudent to evaluate further the relation between the maternal-fetal MBL2 exon 1 genotype and term- preterm births in our population. Genomic DNA was isolated from maternal and cord blood samples of 100 term (38,8±1,2 weeks) and 83 preterm (30,2±2,7 weeks ) deliveries. MBL2 codon 52 (db SNP ID rs5030737), 54 (db SNP ID rs1800450) and 57 (db SNP ID rs1800451) genotyping was performed by PCR-RFLP. The frequency of codon 57 polymorphism was found to be zero for all groups. Maternal codon 54 230 G/A genotype frequency was significantly high in term births than the preterms (p<0.045). In preterm group, the frequency of fetal-maternal GG genotype was also higher than term fetal-maternal group (p<0.001). Pregnancy is an inflammatory process. Therefore, the rise in proinflammatory cytokine level may shorten the gestational process. Previously it was reported that codon 54 230 G/A genotype syntheses MBL2 in low level. As a result, the genotype associated with longer gestational period is an advantage for term pregnancy. The pathological mechanisms of multi factorial disease can not be predicted by using a single marker. There it necessitates further investigation by using other genes enrolling in the delicate balance between fetal-maternal immune system. Therefore the collective data may provide insights of the pathological mechanism and hence may predict the individuals under risk.Item Over kanseri hücrelerinde DNA hasarı ve golgi stresine yanıtta apoptotik mekanizmaların araştırılması(Başkent Üniversitesi Sağlık Bilimleri Enstitüsü, 2024) Aka, Yeliz; Ataç, F. BelginOver kanseri, metastatik aşamada tanı konması ve gelişen kemoterapi direnci nedeniyle tüm jinekolojik kanserler arasında mortalite oranı en yüksek olan kanser türüdür. Tümörün cerrahi olarak çıkarılmasına ve platin bazlı kemoterapiye rağmen, birçok over kanseri hastası kanser tekrarlaması nedeniyle hastalığa yenik düşmektedir. Bu nedenle, over kanseri hastalarının prognozunu iyileştirmek için yeni tedavi stratejileri esastır. Replikasyon stresi over kanserinin yaygın bir özelliğidir ve ATR/CHK1 sinyallemesi hücreleri replikasyon stresine karşı korumada öncelikli olarak rol oynar. ATR inhibitörünün tekil uygulamaları olmakla birlikte kombine tedavi uygulamalar da spesifik hedefler olarak etkinlik gösterirler. Over kanserlerinde ATR inhibitörleri birçok çalışmada kombine olarak mitokondriyal hücre ölüm yolaklarını aktive etse de çalışmalar kısıtlı kalmaktadır. Bu tez çalışmasıyla over kanseri hücrelerinde etkinlik gösterebilecek yeni bir moleküler kombinasyon tedavisinin, hem de bu kombinatoryal uygulamanın moleküler çalışma mekanizmalarının bulunması hedeflenmiştir. Yüksek verimli küçük molekül tarama analizini kullanarak, over kanseri hücrelerinde en etkili hücre ölümü indükleyen kombinasyon olarak AZD6738 (ATR inhibitörü) /Brefeldin A (Golgi stres indükleyicisi) belirlenmiştir. Deneysel çalışmalar ile AZD6738/Brefeldin A'nın eş zamanlı uygulanması, SKOV3 (***p<0.001, kontrol vs AZD6738+BFA) ve IGROV1 (**p<0.01, kontrol vs AZD6738+BFA) over kanseri hücrelerinde kaspaz-3 ve kaspaz-9 aktivasyonu ile apoptotik hücre ölümüne neden olmaktadır. AZD6738/Brefeldin A maruziyeti ayrıca sitozole sitokrom c translokasyonunu da indükler; bu, mitokondriyal hücre ölüm yolunun karakteristik bir özelliğidir. Buradaki en önemli moleküler süreç, SKOV3 ve IGROV1 hücrelerinde AZD6738/Brefeldin A'nın kombinatoryal kullanımı, anti-apoptotik BCL-2 ailesi üyeleri BCL-2, BCL-XL ve MCL1'in ifade seviyelerinde azalmaya yol açarken, özellikle BIM ve PUMA olmak üzere pro-apoptotik BCL-2 proteinlerinin ifade seviyelerinde artışa neden olur. Tüm bu bulgular, AZD6738/Brefeldin A'nın over kanseri için potansiyel bir tedavi yaklaşımı olduğunu düşündürmektedir. Ovarian cancer has the highest mortality rate among all gynecologic cancers due to its diagnosis at the metastatic stage and developing chemotherapy resistance. Despite surgical removal of the tumor and platinum-based chemotherapy, many ovarian cancer patients succumb to the disease due to cancer recurrence. Therefore, new treatment strategies are essential to improve the prognosis of ovarian cancer patients. Replication stress is a common feature of ovarian cancer, and ATR/CHK1 signaling plays a primary role in protecting cells from replication stress. Although ATR inhibitors have single applications, combination therapeutic applications also show efficacy as specific targets. Although ATR inhibitors activate mitochondrial cell death pathways in combination in many studies in ovarian cancer, studies remain limited. This thesis study aimed to find a new molecular combination therapy that could be effective in ovarian cancer cells, as well as the molecular working mechanisms of this combinatorial application. Using high-throughput small molecule screening analysis, AZD6738 (ATR inhibitor)/Brefeldin A (Golgi stress inducer) was determined as the most effective cell deathinducing combination in ovarian cancer cells. Experimental studies have shown that simultaneous application of AZD6738/Brefeldin A causes apoptotic cell death in SKOV3 (***p<0.001, control vs AZD6738+BFA) and IGROV1 (**p<0.01, control vs AZD6738+BFA) ovarian cancer cells, together with caspase-3 and caspase-9 activation. AZD6738/Brefeldin A exposure also induces cytochrome c translocation to the cytosol, which is a characteristic feature of the mitochondrial cell death pathway. The most important molecular process here is that the combinatorial use of AZD6738/Brefeldin A in SKOV3 and IGROV1 cells leads to a decrease in the expression levels of anti-apoptotic BCL-2 family members BCL-2, BCL-XL and MCL1, while it leads to an increase in the expression levels of pro-apoptotic BCL-2 proteins, especially BIM and PUMA. All these findings suggest that AZD6738/Brefeldin A is a potential therapeutic approach for ovarian cancer.Item β-Hücre golgi cisimciliğinin glukolipotoksisiteye stres yanıtı(Başkent Üniversitesi Sağlık Bilimleri Enstitüsü, 2019) Başçıl Tütüncü, Neslihan; Ataç, F. BelginTip 2 Diabetes Mellitus (Tip2DM) etyopatogenezinde genetik ve çevresel faktörlerin rol oynadığı önemli bir hastalıktır. Tip2DM ortaya çıkması için pankreas β-hücre sayısında ve/veya fonksiyonunda yetersizlik gelişmesi gerekmektedir. β-hücre kaybına neden olan en önemli faktör glukolipotoksisite ve buna bağlı gelişen endoplazmik retikulum stresidir. β- hücre gibi sekretuvar hücrelerde, endoplazmik retikuluma yansıyan her stresin Golgi cisimciğinde de hissedilmesi beklenmektedir. Endoplazmik retikulumdan gelen sekretuvar proteinlerin ileri modifikasyonu, veziküler transportu ve sekresyonunu sürdürebilmek için Golgi cisimciğinde de adaptif veya maladaptif moleküler değişikliklerin gerçekleşmesi tahmin edilmektedir. Daha önce yapılmış çalışmalarda, Golgi cisimciğinde fonksiyon kaybına neden olan toksik kimyasalların (Brefeldin A, Golgicide A, vb) bu organeli strese sokarak bir takım adaptif stres cevabı başlattıkları ve bu süreçte spesifik birtakım proteinlerin (ARF4, CREB3/TFE ve HSP 47 gibi) ifadelenmesinin ve/veya aktivitesinin arttığı gösterilmiştir. Bu kapsamda bu çalışmada INS-1E rat β-hücre hattı kullanarak hücrelerde palmitat, glukoz ve glukoz +palmitat ile lipotoksisite/glukotoksisite/glukolipotoksisite oluşturarak Golgi stresi yaratıldı. MTT testi ile hücrelerin canlılık oranları bakıldı. Daha sonra metabolik stres altındaki hücrelerde literatürde tanımlanmış stres proteinlerinin transkript düzeyinde ifadelenmesi incelendi. Her koşulda 8.,16., 24. ve 48. saatlerde Golgi cisimciği yapısal genlerinin, glikozilasyon genlerinin, ARF1, HSP47, CREB3 ve ARF4 genlerinin ifadelenme kat artışı değerlendirildi. Sonuç olarak bu çalışmada INS-1E rat β-hücrelerinde lipotoksisite/glukotoksisite/glukolipotoksisite sonrası ilk saatlerde Golgi cisimciği glikozilasyon enzimlerinden st3gal1 ifadelenmesinin ve golcisidlere cevapta önemli rol aldığı bilinen HSP47 (Serpin1)’in ifadelenmesinin arttığı tesbit edilmiştir. Metabolik toksik ortam uygulamasının 48.saatinde yani hücre canlılığının azaldığı geç saatlerde, CREB3 ifadelenmesinin anlamlı olarak arttığı gösterilmiştir. Bu çalışma literatürde β-hücrelerinde glukolipotoksisitenin golgi cisimciğine etkilerini irdeleyen ilk çalışmadır. Çalışmamızın daha ileri çalışmalara ışık tutacağı düşünülmektedir. Type 2 Diabetes Mellitus is an important disease with diverse polygenetic and enviromental factors in etiopathogenesis. Over the past decades, the central role of pancreatic β-cell dysfunction and loss have become increasingly appreciated. Glucolipotoxicity and its contribution to endoplasmic reticulum stress has been postulated to the worsening β-cell function and survival. In cells with high capacity of secretory function, work load on ER is assumed to be reflected to Golgi apparatus. Factors leading to ER stress is expected to converge adaptive and/or maladaptive changes in Golgi structural proteins, enzymes and secretory function. In this regard, there are documented Golgi stress proteins (TFE , HSP 47 CREB3/Luman, and ARF4,) of which genetic expression and/or activity have been increased after derangement of Golgi function with golcisides like Brefeldin A, Golciside A etc. We hypothesized that glucolipotoxicity in diabetes can trigger a Golgi stress response in pancreatic β-cells. In order to test a Golgi stress response in β-cells of a diabetic metabolic millue, we tried to create lipotoxicity/glucotoxcitiy/glucolipotoxicity in INS-1E rat β-cells with palmitic acid, glucose and both together and investigated the expression of the genes documented as Golgi stress proteins before in the literature. In this context palmitic acid, glucose and both together were applied to INS-1E rat β-cells and analysed at 8th, 16th, 24th and 48th hours. MTT assay was used for cell viability analysis. Expression rates of Golgi structural proteins, Golgi glycosylation enzymes, ARF1, HSP47, CREB3 ve ARF4 proteins were measured in all experimental conditions. In our study we determined an increase in expression levels of Golgi glycosylation enzyme stgal1 and HSP47 in the 8th hour of all experimental conditions. In the 48th hour, when the viability of most cells were lost, expression rate of CREB3 was found to be increased. Our study is the first one in English literature which investigated Golgi stress response in β- cells under lipotoxicity/glucotoxicity/glucolipotoxicity. HSP47 and CREB3 were found to be important mediators of Golgi stress response in β-cells under metabolic stress. In this regard, this study will form basis for further studies dealing with Golgi stress in diabetes mellitus.
